Y-STR Testing

Genetic markers, DNA Short Tandem Repeat (STR) in the Y chromosome can be used to determine if there is a genetic paternal relationship between two males. This test can be conducted not only between two brothers but also with several family members from previous generations such as grandfather or/and uncles from the same paternal lineage.

What is Y-STR testing?

Forensic Kinship DNA testing fundamentally involves comparing the DNA profile of the questioned individual with the DNA profile of other individuals from the same family lineage. In certain conditions where samples from biological parents or children are unavailable (either due to parental death or in cases of mass disasters), then kinship testing can be conducted. 

DNA kinship testing using Y chromosome STR markers could provide scientific evidence for clarifying paternal lineage by comparing DNA profiles between two (or more) male individuals. 

The Y chromosome is only possessed by males, thus its inheritance is only passed in the paternal lineage. A son inherited the Y chromosome only from his biological father. Therefore, the DNA profile of the Y chromosome of the biological father is the same with the DNA profile of the Y chromosome of his male child (son).

The Y-STR testing at Genos Laboratory uses 23 allele genetic markers of Y chromosome-STR. If the entire set of markers (23 alleles) on the Y chromosome between the individuals being compared is matched, it can be concluded that the tested individual originates from the same paternal lineage. We can also confirm with 100% accuracy that if the Y chromosome patterns between 2 individuals are not the same (do not match), the two males are not related from the same paternal lineage.  

Generally, the sample used is blood. Blood samples are taken from the vein using a sterile syringe needle with volume less than 1 mL or from the fingertip pricked with a sterile needle (finger prick). Another sample option can be used is buccal swabs collected by gently swabbing the inside of the cheek with a cotton swab. The type of sample does not affect the DNA test results.

At least two (2) male individuals or brothers whose paternal lineage is doubtful. Comparison of Y-STR patterns can also be carried out on a child with his previous generation family members, such as grandfather and/or uncles from the same paternal lineage.

Fasting from food and beverage is not necessary

  • Personal identification card (ID card/Passport) Birth certificate is needed for individuals under 17 years old
  • Individuals under 21 years old must be accompanied by a Legal guardian
  • Recent colored photo size 3x4 cm
  • No referral letter from a doctor is required

The results of the Y-STR kinship testing from the Genos Laboratory can be used for family reunions, identification of a man in cases of mass disasters, and the search for missing sons (families).

Genos Laboratory is highly concerned about the chain of custody, to ensure that our DNA test results can be legally accountable. We have standardized operating procedures (SOP) aimed at maintaining the chain of custody. The chronology of sample collection to the release of DNA test results is well documented, including the interactions with the samples. Every individual/parties involved in DNA paternity testing is required to sign a DNA examination informed consent and DNA requisition form on a duty stamp, consciously and without coercion. Minors would be represented by a legal guardian.

Y-STR testing carried out at the Genos Laboratory takes 5 working days from the day one (the confirmation day of sample received).
